I'm pretty darn sure that Esau and Jacob were foreshadowed all the way back in the first episode. Esau and Jacob were dressed in Black and White, respectively, in The Incident's teaser. In the pilot, Locke was explaining backgammon to Walt, saying "Two players, one dark, one light." This seems to me to indicate that the entire show is a game being played by Esau and Jacob.
